This is not possible without some customization. One way to do it would be to remove the regular Location field from all template files (so it is not displayed anywhere) and add a new custom profile field to contain the same information. This you can then set to be required at registration. You might also want to search for a MOD once the .com site is back online.
Why do the standard profile fields not have the same flexibility as custom profile fields? Because custom profile fields were a later addition to the software and replacing the standard profile fields with out of the box configuration for the custom profile field feature was postponed to a later version.
